Will the volumite be able to control the volume directly on multiple Buffalo modules, or through multiple Joshua Trees, as long as no balance control is required?

Yes, it will be able to control buffalo (multiple is fine, but they all will have he same level) and Joshua Tree via the I2C relay driver (same applies).

I am planning to build DAC based on a Buffalo-II/IVY-III Dual-Mono design. I will also be using the S/PDIF (CS8416) Receiver/MUX Module, TOSLINK Optical Input and USB Receiver Module. My question is what approach will give the best result for volume control, the Volumite or Joshua Tree? Any input most appreciated.

First choice would be Volumite for sure. But either approach is fine.
