Buff III was locked onto a MiniDSP USB interface but now has lost lock. Lock light flashed regularly. What does this mean anyone?

That it tries to lock, getting locked and quits again - I guess.

Try another source if you have one.

Is this I2S or S/PDIF?

I have seen this in a few scenarios:

Bad solder joint to one of the 3.3V Tridents
A loose I2S wire, or bad solder joint
I2S Wiring (long wires or noise injected from I2S lines into a crossing power supply line).

Thanks for your suggestions Brian, but none of these are issues as far as I can see. I have shortened the I2S wires as much as physically possible - made no difference. The lock dropouts increase with sample rate increase BTW.

I had a similar issue with the USB ministreamer that could also not be resolved (it would randomly lose lock). I tried multiple ministreamers, all with the same issue. Eventually I swapped it for a DIYinHK USB receiver which worked flawlessly.

Perhaps I am flogging a dead horse here and there is some fundamental incompatibility. Are there any known multichannel/surround sound USB devices that are known to work with the Buffalo?
